Pharmacodynamics and pharmacokinetics

Pharmacodynamics

Lomflox is a synthetic antimicrobial agent, a representative of difluoroquinolones. The peculiarity of its structure significantly improves the pharmacokinetic properties of the preparation and protects the molecule from biotransformation in the body. Has a wide range of bactericidal action. It breaks the replication of the microbial, blocking the DNA-gyrase of the bacteria, which leads to irreversible damage to the intracellular synthesis of the pathogen and leads to its death.
It is active against gram-positive and gram-negative microorganisms: gram-positive aerobes, gram-negative aerobes, chlamydia, mycoplasma (some strains), uroplasm, legionella. It acts on microbes that are insensitive to the action of aminoglycosides, penicillins and cephalosporins. Has a pronounced postantibiotic effect. Resistance of microorganisms develops slowly.
Of all the fluoroquinolones, ciprofloxacin, ofloxacin and lomefloxacin suppress the growth of mycobacteria tuberculosis. Lomflox refers to reserve drugs for the treatment of tuberculosis, it is used only as part of combination therapy.
Anaerobes, streptococci (pneumoniae, groups A, B, D, G), Ureaplasmaurealyticum, Pseudomonascepacia, Mycoplasmahominis are resistant to Lomefloxacin.

Pharmacokinetics

All fluoroquinolones are rapidly absorbed from the digestive tract, but best of all are ofloxacin, pefloxacin and lomefloxacin. Smah marks in 60-90 minutes. It is slightly bound by blood proteins and has a good penetrating ability with maximum concentration in various tissues and intracellular spaces.
T 1/2 is 7 hours, that is, elimination from the blood is slow. A long half-life and the presence of a postantibiotic effect make it possible to prescribe the drug once a day. The main way of excretion through the kidneys (up to 70%). In 12 hours, 53% of the dose is taken. With renal failure, it is necessary to correct the dose.

Instructions for use Lomflox (Method and dosage)

Tablets are taken orally 400 mg 1 time / day. Their reception does not depend on the time of food intake. If the kidney function is disturbed, the initial dose is 400 mg, with a transition to 200 mg per day. With cirrhosis of the liver, there is no need to adjust the dosing regimen, provided that the kidney function is not impaired.
The duration of the course is determined by the doctor and depends on the severity of the disease: from 3 days (with uncomplicated urinary tract infection and uncomplicated gonorrhea) to 28 days (with tuberculosis).
Instructions for use Lomflox contains a warning that during the period of treatment you should avoid exposure to the sun. The risk of a photochemical reaction is reduced if taking the drug in the evening.

Overdose

It is manifested by symptoms: tremor, hypersalivation, nausea, vomiting, depression, decreased activity, respiratory failure, convulsions.
Treatment: gastric lavage, sorbent intake, symptomatic therapy, rehydration.
Hemodialysis is ineffective.

Interaction

Lomflox is an antagonist of Rifampicin, and therefore, their combined use in the treatment of tuberculosis is not recommended. It is acceptable to use together with isoniazid, streptomycin, pyrazinamide.
Lomefloxacin increases the activity of anticoagulants, and increases the toxicity of NSAIDs.
There is no cross-resistance with cephalosporins, metronidazole, penicillins, aminoglycosides and co-trimoxazole.
Probenecid slows the excretion of lomefloxacin by the kidneys.
Antacids, sucralfate and other drugs that contain iron, magnesium, and aluminum, slow the absorption of the drug and reduce its bioavailability.
Drugs that block tubular secretion, significantly slowing the withdrawal of this drug.
Do not use the drug at the same time as alcohol.

Storage conditions

At temperatures up to 25 ° C.
Shelf life - 3 years.

Reviews of Lomflox

Often there are Reviews of Lomflox, which was used for the treatment of mycoplasmosis and ureaplasmosis. Effective was the simultaneous use of this drug and Doxycycline, the treatment was supplemented with immunomodulators, antifungal agents and Wobenzym.
In chlamydial vaginitis, colpitis, urethritis antibacterial agents of 3 pharmacological groups were assigned: tetracyclines (Doxycycline), macrolides (Sumamed, Klacid, Vilprofen), fluoroquinolones (Lomflox). Patterns were used in which antibacterial agents were used consistently.
It is reported about the effectiveness of the drug in the treatment of prostatitis, but even in this case there was an integrated approach to the treatment of the disease, so do not rely on advice from visitors to the forums, and on treatment issues, contact a specialist.
In the event that the drug is ineffective, there is one answer - an inadequate survey was conducted. In all cases of bacterial infections, it is necessary to determine the type of pathogen and it is necessary to establish its sensitivity to fluoroquinolone (or antibiotic).
Among the side effects, many patients note nausea, dryness and bitterness in the mouth and upset of the stool during the drug intake. Often there are complaints of dizziness, headache and retardation.
