pharmachologic effect

Contraceptive drug of combined action for oral administration. Helps to reduce the secretory function of the pituitary gland, responsible for the production of gonadotropic hormones, inhibits the process of maturation of follicles, prevents ovulation. Ethinyletradiol (a synthetic analogue of estradiol), an estrogenic component of Lindynette 30, is a regulator of the menstrual cycle. Gestodene (gestagen component), one of the most active synthetic gestagens, by activity and selectivity exceeds the effect of the natural yellow body. It is used in minimal dosages, which practically do not affect the fat and carbohydrate metabolism, does not show androgenic properties.
Lindynette 30 contributes to changing the nature of cervical mucus, preventing sperm from entering the uterus, reduces the sensitivity of the endometrium to implantation of the blastocyst (the initial stage of development of the embryo), has a beneficial effect on the course of the menstrual cycle. With regular use of Lindynette 30, the volume of blood loss decreases, the incidence of ectopic pregnancy, dysmenorrhea, functional ovarian cysts, fibrous tumors in the mammary glands, fibroadenomas, congestion and inflammations of the pelvic organs decreases, and the condition of the skin in case of acne is normal.

Pharmacokinetics

Gestoden. Rapidly absorbed when taken orally. Сmax in the blood plasma after the application of a single dose of the drug after 60 minutes. makes 2-4 ng / ml. Bioavailability - 99%. It interacts with albumin and globulins that specifically bind sex hormones (70-75%). 1-2% of gestodene is in a free state. The average plasma clearance is 0.8-1 ml / min / kg. Half-life is in the range of 12-20 hours. Excreted in the form of metabolites 40% - through the intestines, 60% - with urine. Under the influence of an active component, the concentration of SHBG (sex hormone binding globulin) increases 3 times. With daily use of Lindynette 30, the level of gestodene in plasma increases by 3-4 times, reaching saturation in the second half of the menstrual cycle.
Ethinyl Ethiol With internal intake, it is quickly and almost completely absorbed. 1-2 hours after the application of Cmax in the blood is 30-80 pg / ml. Absolute bio-cost - 60%. About 98.5% of the active ingredient binds non-specifically to albumin, increasing the level of SHBG. The average volume of distribution is 5-18 l / kg. Almost completely metabolized in the liver. Plasma clearance is 5-13 ml / min / kg. Excreted together with bile and urine (in the form of metabolites) in the ratio 3: 2. Half-life is 24 hours. A stable concentration of ethinyletradiol is established after 3-4 days of intake. At the same time, a 20% increase in the level of the active component is observed (in comparison with taking a single dose).

The duration of use is 21 days, then a week break (during this period menstrual-like bleeding occurs). The next course starts on the same day of the week from which the previous one started.
In that case, if the drug is started after using another oral contraceptive, the first pill is taken on the first day of the menstrual bleeding, or the next day after taking the previous drug. The exceptions are mini-pills (hormones containing progestogen only). In this situation, the transition to Lindynette 30 is allowed on any day of the menstrual cycle.
After a medical abortion (in the first trimester of pregnancy), the drug is started to be used immediately, it is not advisable to use additional contraceptives in this situation. Contraceptive use after childbirth or abortion, carried out in the second trimester of pregnancy, should begin after 28 days. In this case, the first week of applying additional methods of contraception.
With a single skip drug pill must be taken as soon as possible. If no more than 12 hours have passed since the last dose, the effect of Lindynette 30 is not reduced, and additional contraceptive methods are not required. With a break of over 12 hours, the missed pill can not be taken, and the next - in normal mode. However, it should be borne in mind that the effectiveness of the drug may be reduced. Therefore, during the week it is recommended to use additional means of protection from unwanted pregnancy.
If diarrhea or vomiting develops after the use of a hormonal agent within the first 2-4 hours, you should act in the same way as in case of skipping the drug.
Note: Lindynette 30 is able to change the date of the beginning of the menstrual cycle. To delay the monthly it is recommended not to take a break between using the previous and next pack.

Drug interaction

Antibacterial drugs (rizeofulvin, ampicillin, rifampicin) and barbiturates reduce the effectiveness of the contraceptive by changing the intestinal microflora. With simultaneous use, there is a risk of breakthrough bleeding (the appearance of bleeding between periods). If it is necessary to use antibiotics for the entire course of etiotropic therapy and another week after treatment (for rifampicin - 4 weeks), it is recommended to use additional contraception.
Increase the bioavailability of the drug inhibitors of liver enzymes and ascorbic acid.
