Pharmacodynamics and pharmacokinetics

Information about Trigan-D in Wikipedia is missing, and the interested category of people have questions, "Trigan-D, what is this?" Or "What is Trigan-D?"
The first component is paracetamol: it has antipyretic, analgesic and weak anti-inflammatory effect. Oppresses cyclooxygenase of the first and second type in the nervous system and other tissues, as a result, stops the synthesis of prostaglandins - mediators of pain, regulation of body temperature and inflammation.
The second component is dicycroverine hydrochloride: it possesses a small non-selective m-holin-blocking and myotropic antispasmodic effect on smooth muscle cells of internal organs. In therapeutic doses has a relaxing effect on smooth muscles, not accompanied by atropine-like side effects.
The combined use of these two components makes it possible to relax the smooth muscle tissues of the internal organs and to arrest sensations of pain.

Pharmacokinetics

Well absorbed from the intestine. The maximum concentration in the blood is fixed after one hour. Paracetamol is transformed in the liver with the formation of a number of metabolites, one of them N-acetyl-benzoquinomine can be damaging to act on the kidneys and liver. 80% of the drug is excreted by the kidneys and in small amounts with feces.

    spasm of smooth internal organs;
    algodismenorea;
    hepatic, intestinal or renal colic;
    neuralgia;
    toothache or headache;
    myalgia;
    fever in infectious and inflammatory diseases.

    Sensitization to paracetamol or dicycoverin.
    Gastric or duodenal ulcer.
    Bowel diseases complicated by obstruction.
    Hypovolemic shock.
    Reflux esophagitis.
    Myasthenia gravis.

The components of the drug in patients with hepatic or renal impairment, deficiency of glucose-6-phosphate dehydrogenase, glaucoma, blood diseases, hyperbilirubinemia, viral hepatitis, alcoholism and in old age can worsen the course of these diseases, from which the Trigan-D tablets in this category of persons should be used with extreme caution.

    Allergic reactions: Quincke's edema, rash, hives, Stevens-Johnson syndrome, itching, Lyell's syndrome.
    Reactions from the hormonal system: hypoglycemia.
    Reactions from the excretory system: interstitial nephritis, urine retention, pyuria, papillary necrosis, potency reduction.
    Reactions from the hematopoietic system: methemoglobinemia, hemolytic anemia.
    Reactions from vision: mydriasis, accommodation paralysis, blurred vision, increased intraocular pressure.

Tablets Trigan-D, instructions for use (Method and dosage)

Instructions for use on Trigan-D recommends the use of the drug inside 1 tablet up to three times a day. The largest single dose is two tablets, the daytime - four tablets. Duration of intake - no more than five days at use, as an analgesic and three days - as an antipyretic.
Do not advise to exceed the daily dose, as an overdose of the drug causes liver failure, for which Trigan-D (if necessary to increase the dose or increase the standard duration of treatment) should be used under the supervision of a doctor.

Overdose

Overdose Trigan-D, symptoms: tachypnea, tachycardia, fever, seizures, agitation, pain in the stomach, appetite reduction, thrombocytopenia, anemia, nephrotoxicity, hemolytic or aplastic anemia, pancytopenia, methemoglobinemia, hepatonecrosis.
Therapy: stop the use of the drug, perform gastric lavage, take adsorbents, stimulate the formation of glutathione (for example, acetylcysteine intravenously) and activate conjugation (methionine).

Interaction

Effects of dicycloverin stimulate amantadine, antipsychotic drugs, first-line antiarrhythmics, benzodiazepines, narcotic painkillers, MAO inhibitors, sympathomimetics, nitrates, nitrites, tricyclic antidepressants.
Dicycloverin increases the content of dioxin in the blood.
Amplifiers of microsomal oxidation (ethanol, barbiturates, rifampicin, tricyclic antidepressants) stimulate the synthesis of hydroxylated metabolites in the liver, this can lead to the development of strong intoxications, even with weak overdoses of paracetamol.
Inhibitors of microsomal oxidation reduce the possibility of the appearance of a hepatotoxic effect.
Adrenostimulators and anticholinergic drugs increase the risk of unwanted effects.
Paracetamol activates the action of indirect anticoagulants.

Reviews about Trigan-D

As evidenced by the reviews, the drug helps to effectively cope with spasms and pain in the vast majority of cases.
Also known is the narcotic effect of Trigan-D and its use as a drug by purification from paracetamol. As a result, after using the substance obtained, side effects occur on the part of the psyche and the nervous system, called "arrival". You should know that this is an extremely harmful effect, which can lead to irreparable consequences. At specialized forums, there are often questions such as "how to beat off paracetamol from Trigan to get a drug?". We strongly advise not to engage in such experiments with drugs and your body.
