Pharmacodynamics and pharmacokinetics

What is nitrofural?

Nitrofural is a derivative of nitrofuran, which is yellow or yellow with a greenish tinge, a bitter-tasting, fine-crystalline powder.
The powder is almost insoluble in ether, slightly soluble in ethanol and water, but readily soluble in alkali solutions.
The gross formula of nitrofural is C6H6N4O4.
Wikipedia indicates that one of the properties of nitrofuran compounds is photosensitivity. Therefore, the diluted solution should not be exposed to daylight. Particularly harmful for solutions of furacilin UV rays, which deeply and irreversibly damage the molecule of nitrofural.

Pharmacodynamics

The properties of nitrofurans are due to the presence in their molecule of an aromatic nitro group, which is similar to the chloramphenicol (levomycetin) present in the molecule.
The mechanism of action of nitrofural is different from the mechanism of action of other chemotherapeutic drugs: under the influence of nitrofural from proteins of microbial cells (flavoproteins), amino derivatives that are highly reactive are formed, which have the ability to change the conformation of macromolecules (including ribosomal proteins).
As a result, the tertiary and quaternary structure of the protein is deformed, the flow of metabolic processes in the microbial cell is disrupted, and the cell dies.
The substance increases the absorption capacity of the macrophagic (reticuloendothelial) system and promotes the enhancement of phagocytosis.
Nitrofural is effective against Gram (+) and Gram (-) bacteria, as well as against some protozoa and viruses. Sensitive to the drug are: Shigella (Sh. Dysenteriae, flexneri, boydii, sonnei), Escherichia coli, Streptococcus, Staphylococcus, Salmonella, Clostridium Perfringence.
Resistance to nitrofural develops slowly, while it does not reach a high degree. The drug is effective in the resistance of pathogenic microflora in other antimicrobial drugs (which are not derivatives of nitrofuran).
Nevertheless, in the hospitals there are completely resistant to nitrofural strains of microorganisms.

Pharmacokinetics

With external and local application nitrofural is absorbed in very small amounts. The substance penetrates through the histiocytic barriers and is evenly distributed in the physiological fluids and tissues of the body.
Metabolized nitrofural mainly by reducing the nitro group. The excretion of metabolic products is carried out by the kidneys, in addition, in part they are ex- tracted with bile.
