Composition

One tablet of Ganaton contains, as active substance, the hydrochloride of tautopride in an amount of 50 mg. As auxiliary substances contain: corn starch - 15 mg, lactose - 34.975 mg, carmellose - 20 mg, silicon anhydrous acid - 4 mg, magnesium stearate - 1 mg, hypromellose - 4.4 mg, titanium dioxide - 0.2 mg, macrogol 6000 - 0.4 mg, carnauba wax - 0.025 mg.

Form of issue

The drug is available in tablets coated with a light colored coating, rounded, with a strip on one edge and the inscription "HC 803". Seven, ten or fourteen such tablets in a blister. Five, four, three, two or one such blister in a cardboard box.

Mechanism of action. Etymopolid hydrochloride activates gastric motility by suppressing D2-dopamine receptors and acetylcholinesterase. Itopride inhibits the destruction of acetylcholine and enhances its release.
Tetropride hydrochloride also has an antiemetic effect by inhibiting the D2 receptors localized in the trigger region and causes a dose-dependent depression of emesis caused by apomorphine.
The drug activates the propulsive motility of the stomach and accelerates the transit of food through the stomach and its emptying. Does not affect the concentration of gastrin in the blood serum.

Pharmacokinetics

Suction. The drug is quickly and almost completely absorbed from the stomach and intestines. Bioavailability is 60% - this is due to the transformation of the molecule during the first passage through the liver tissue. Eating does not affect bioavailability.
After ingestion of 50 mg of aseptide hydrochloride, the maximum concentration is achieved in half an hour. In the following doses of the drug from 50 to 200 mg three times a day for a week, the pharmacokinetics of the drug itself and its derivatives did not change, and the cumulation effect was not observed.
Itopride binds to plasma proteins (albumins, to a lesser extent with alpha-1-acid glycoproteins) by 96%. Actively carried on the tissues and detected in large concentrations in the small intestine, kidneys, adrenal glands, liver and stomach. Penetrates into the dorsal and the brain in small amounts. Capable of penetrating into breast milk.
Metabolism. The drug undergoes an active transformation in the liver. Three metabolites are known, only one of them exhibits weak activity, which does not have much significance.
Itopride is decomposed by flavine-dependent monooxygenase, its concentration and efficacy in humans depends on congenital polymorphism. In patients with trimethylaminuria, the half-life of the isotope increases.
Excretion. The drug and its metabolites are excreted in the urine. The terminal half-life of the endopride is 6 hours.

Indications for the use of Ganaton

The drug is used to treat functional dyspepsia of non-ulcerative nature (chronic gastritis). The use of the drug is justified for cupping:
    sensations of rapid saturation;
    bloating;
    discomfort and pain in the upper abdomen floor;
    anorexia;
    nausea;
    heartburn;
    vomiting.

Contraindications

    Gastric or intestinal bleeding, mechanical obstruction or perforation of the gastrointestinal tract.
    The period of breastfeeding (lactation).
    Pregnancy.
    Children under the age of 16 years.
    Sensitization to taepride or other component of the drug.

Side effects

Instructions for using Ganaton recommend taking this medication internally at 50 mg (1 tablet) three times daily before meals. The recommended daily dose of the drug is 3 tablets. This dose can be reduced based on the age of the patient.

Overdose

There are no reports of overdose. If it does, it is necessary to rinse the stomach and begin symptomatic treatment.

Interaction

With the simultaneous administration of ganaton with diazepam, warfarin, diclofenac, ticlopidine hydrochloride, nifedipine and nicardipine hydrochloride, there are no disturbances in the interaction between itopride and blood proteins.
Itopride improves gastric motility, as a result of which it affects the absorption of other substances simultaneously used with Ganaton. Particular care should be taken when using drugs with a small therapeutic index, prolonged release of the active ingredient and enteric coating.
Anticholinergic drugs weaken the effect of itopride.
