Pharmacodynamics

Prokinetics, which has an effect on the motor function of the gastrointestinal tract. Prevents the anti-peristaltic contractions of the musculature. Has a combined mechanism of action, which consists in interacting with D2-dopamine receptors and blocking acetylcholinesterase. Prevents the destruction of acetylcholine and activates its release. This causes an increase in gastrointestinal motility throughout the digestive canal: accelerates the passage of food through the stomach and intestines, improves their emptying.
The antiemetic effect is related to the interaction of tautopride with D2 receptors, which are located in the trigger zone (the bottom of the fourth ventricle of the brain). Suppresses vomiting, which is caused by apomorphine and this effect is dose-dependent.
The advantage is the low ability to penetrate the BBB, therefore there are no extrapyramidal and neuroendocrine effects. Does not change the concentration of the hormone gastrin in the serum.

Pharmacokinetics

Quickly absorbed in the digestive tract. Bioavailability of 60%. Food does not affect the bioavailability of the drug. The maximum concentration is 0.5 hours. With repeated administration for 7 days, cumulation is minimal.
96% bound to proteins. It is well distributed in tissues and is found in high concentrations in the small intestine, stomach, kidneys, liver and adrenal glands. Slightly penetrates the brain.
Metabolized in the liver to 3 metabolites, one shows little activity. Itopride has no effect on CYP2C19 and CYP2E1. The elimination half-life is 6 hours. It is excreted by the kidneys.
