Pharmacodynamics and pharmacokinetics

Terazosin is an α1-adrenoblocker that is able to selectively block α1-adrenergic receptors in the smooth muscles of celiac vessels, prostatic vessels, and the neck of the bladder. As an antihypertensive drug expands large resistance vessels and reduces OPSS, activates the system of RAS (renin-angiotensin-aldosterone).
Relaxation of the smooth muscles of the neck of the bladder and contractile elements of the prostate, help reduce the dysuric phenomenon, for example, with adenoma. Due to a decrease in the level of total cholesterol, triglycerides, an increase in the level of HDL is normalized lipid profile in plasma. Long-term therapy leads to regression of left ventricular hypertrophy.

The pharmacokinetics of Terazosin

After ingestion, Terazosin undergoes rapid, almost complete absorption (bioavailability above 90%). Pre-system biotransformation is insignificant, therefore, to achieve maximum plasma concentration, only 1 hour is needed. In plasma, 90-94% of the substance binds to proteins. Metabolism in the liver occurs before the formation of 4 metabolites.
Half-life is no more than 12 hours, about 40% is excreted by the kidneys (up to 10% is unchanged Terazosin), the remaining 60% is through the intestine (more than 50% are biodegradation products). If a patient has liver pathology, then a decrease in clearance and prolongation of pharmacological effects are possible.
